此前,該團隊已通過瞬時激活轉錄因子ETV2和NKX3.1,分別將 iPSC 高效分化為誘導內皮細胞(iEC)和誘導血管壁細胞(iMC),表明了血管類器官中最主要的兩種細胞類型都可以通過激活轉錄因子進行正向編程的方式獲得,這為接下來血管類器官分化新方案的研究打下了堅實基礎。
在這項最新研究中,研究團隊提出了一種通過使用多西環素誘導或 modRNA 調控系統對轉錄因子 ETV2 和 NKX3.1 進行正交激活,從而從誘導多能干細胞(iPSC)生成血管類器官(VO)的簡化方法。
該方法采用轉錄因子ETV2與NKX3.1的正交激活策略,分別誘導iPSC向內皮細胞和血管壁細胞分化,通過預誘導中胚層祖細胞(Mesoderm Progenitor Cell,MePC),再將兩種初步誘導的iPSC按1:1混合并3D培養,在 5 天時間內即可形成直徑約250 μm、結構統一的功能性血管類器官,且無需細胞外基質。這些血管類器官在嵌入細胞外基質后會進一步成熟,形成了更大且結構更完善的血管。
接下來,研究團隊通過單細胞 RNA 測序揭示了這些血管類器官中血管細胞的異質性,不同轉錄因子誘導時長會導致血管譜系偏倚:短時激活ETV2(1天)更易獲得動脈樣iEC,而長時激活(3天)則促進靜脈樣或具更高血管新生潛能的 iEC,在體內移植中形成更多灌注血管。這一可調控的“命運開關”,為未來按需構建特定類型血管網絡提供了工具。
進一步體內實驗表明,將這些血管類器官植入免疫缺陷小鼠體內后,形成了有血液灌注的血管,并在下肢缺血模型和胰島移植模型中促進了血管重建。
該研究的核心發現:
